Natural Retinol Analogs Potentiate the Effects of Retinal on Aged and Photodamaged Skin

Retinal has approximately 3 times higher biological activity than retinol, and its conversion speed to retinoic acid is over 11 times faster, greatly enhancing the skin aging improvement effect.

Because of side effects such as skin damage, erythema, and desquamation caused by high retinoic acid (RA) concentration, RA use is prohibited in cosmetic products in Europe and the US. Instead, endogenous RA precursors like retinal (RAL) and retinol (ROL) are used. To be biologically active, these precursors must first convert to RA. RAL is converted to RA by keratinocytes in a single step, while ROL requires an additional step, using RAL as an intermediate. By bypassing the first of the two rate-limiting oxidation steps, RAL is approximately threefold more biologically active than ROL. Its conversion to RA is estimated to be at least 11 times faster. Moreover, RA transformation from RAL occurs in both differentiating and non-differentiating keratinocytes, unlike ROL which occurs only in differentiating keratinocytes. These data suggest that RA transformation and retinoid activity in cosmetic formulations can be maximized using RAL.
